Do you get the same exact issue from any layout?
Just ran a quick test here on a windows xp system and the button that I copied was set to change fill colors on hover. Pasting the button into other layouts produced a button that also changed color when a mouse havers over it.
Does the button use conditional formatting?
Conditional formatting currently over-rides the 'hover' styles, I wish there was a way to toggle this.

There are no conditional formatting options set on these buttons.
The same button that works on one layout was copied and pasted to others that work, but doesn't work on some. I can even set the color to change when pressed and that works, but not on hover. I can create a new button using the button maker in the tool bar, and it works. But if I use a pre-existing button (that works) from another layout, it doesn't.
Don't know if it matters, but the buttons are from FMP Graphics pack.
If you can copy and paste from one layout and the pasted button works as expected, but copy/pasting it from a different layout does not, it's possible that you have an issue on that layout.
Have you tried recovering the file just to rule out possible file corruption?
And it's not impossible to have a corrupted layout.
Wow! Do I feel stupid. But, at least I figured this out by myself.
There has to be at least one record present for hover to work.
