When FileMaker Pro connects to a database file, it pings the file to make sure there is a connection. If the connection can't be made, FileMaker will continue trying for two minutes. If at that time, the connection still can't be established, the connection is dropped.
Have you monitored your network to make sure the external drive is still connected?
Is there another machine that stays connected to the file? If not, then you have a network issue. If the other machine stays connected, the issue is on the Windows 10 machine.
Thank you for your answer. I checked again and it seems the connection is lost after 2 minutes of inactivity on FileMaker.
I have been using the same file on the same computer with Windows 7 for years without problems before upgrading to Windows 10. I also use the datafile on another Windows 7 computer everyday without any problem. I guess the problem must be with how Windows 10 handle the connection with the USB drive then.

Since Windows 10 has just been released, do you have another internal/second disk drive (e.g., D: drive)? If you place the file on the second disk drive, does the issue still occur? I'd like to report if this is a USB drive issue or a bigger second drive issue.
This is a laptop with a single internal drive, so I won't be able to test this configuration on the same machine unfortunately. I can try to find another Win 10 computer with two internal drive to test but it might take a few days.
I did some testing and the problem only occur with the BUP fast SSD USD drive connection. Everything was fine when the datafile were on a USB key or a regular (not SSD) external harddrive.
Let me know if there is a fix because I would rather use my external SSD drive for datafile storage.
