Would you be able to re-clarify your question? Are you asking if a web browser can access FileMaker Pro without the use of Instant Web Publishing?
If I have upload a database in FileMaker Server.
I want to know whether the FileMaker Pro 9 can open the database from internet(not from LAN) direct.
You can open an FileMaker file that is hosted over the Internet. Either through hosting the file with FileMaker Pro or FileMaker Server. FMS is recommended. The Internet is just a wide-area-network (WAN) so just about anything you can do on a local-area-network (LAN) you can do on a WAN.
To open files or control FMS from outside your LAN you will need to setup the proper ports to be forwarded to your server. (See the link below.) Once you do that you will need to find out what your WAN IP address is. It's important to have a static IP address as a dynamic IP will change and it will be hard to know when you receive a new address. It is possible to use a service that will keep up with your dynamic IP but it's just easier if you have a static IP.
Once you know your static IP address and you are outside your LAN go to open remote and type in fmnet://xxx.xxx.xxx.xxx/your_solution and your in. (As long as you set everything up correctly.) Obviously replace xxx.xxx.xxx.xx with your IP address and your_solution with the name of your solution.
It is slower than being local. Things like sorted portals, auto fill fields and finds will take longer so develop accordingly.
Here is a list of FileMaker ports. Read over it as not necessary to forward all the ports listed.

I have host the database via Filemaker Server, and open Filemaker Pro->File->Remote->fmnet:/WAN IP address: port/database name. But I can not open it. (Database can open via http://WAN IP address: port/fmi/iwp)

File-Manage- Accounts and privileges - Select user - Edit - Edit (privilege set) for this user to access via Filemaker (fmnet) or Web Browser (Instant web pub).

don't forget to map the 5003 port in your router and access on your firewall
bind the traffic from Internet to your FM server with it's local IP (input port can be different when using htt://ip:port)
I done this, but can not access the database via fmnet.

Solved!!! Forget to open port 5003.
