Do you have Filemaker Pro Advanced ?
the html version of the developer design report in fm pro advanced can show where every field is used Except where you use features like set field by name using a calculation Or if you reference them by an object name. If you don't do that then the design report has what you need. Look at all the fields and ones that have nothing in subsequent columns should be good candidates for deletion.

This blog post should be helpful. Where is say "Renaming" think "Deleting". Sometimes we do this in 3 steps. 1. Rename ; 2. Allow time to pass and usage to occur ; 3. Delete or rename as zzz_GlobalFields

If we want to have the advantage we should avoid the disadvantages. We need to determine what types of FileMaker code can break by renaming and what are the causes. Then we can track down code elements and sanitize the code against breaking (for example by using GetFieldName) or coordinate the changes.

I'd start by individuating layouts you don't use any more (not referenced by scripts or buttons) and deleting them. Then DDR like fclark said. Be aware that other DBs can reference layouts and fields in this DB, don't know if it's the case.
