Is there any way or workaround to get Field Validation "Unique value" to distinguish between lowercase and uppercase. That is, a is unique as compared to A or hello is unique as compared Hello.

That does work, but be aware that it carries additional implications. Any searching done on the field will have to be in the proper case, otherwise it won't return the records correctly. So you'll need to be sure your users are aware of this before they get completely frustrated (or get bad results). Your scripts will also need to account for the new indexing.
Thank you Mike for your input. Changing to unicode really did help. In this special case there will be no user search in this field, only by script controlled by myself.
