FileMaker Pro 12 files are in the same format as newer versions (.fmp12) and will definitely open in FileMaker Pro 16 Advanced. It is hard to guarantee you will not have to make any changes to the database. There are some features that have been changed or removed. This might cause a change of behavior in your solution. A database created in FileMaker Pro 12 can be opened with FileMaker Pro 16 (advanced or not). There is no file format change and nothing to convert. Keep in mind that any changes to database with features new in 16 will not work in prior versions. See these charts for comparisons of version changes:

Are you HOSTING the database on a FileMaker Server? There are previous versions of FileMaker Pro that do not work with FMServer 16 and the other way.
