WebDirect multiple machine configuration connection failure

Upgraded a WebDirect multiple machine configuration from FileMaker Server 15.0.3 to FileMaker Server 16.0.4, but cannot get the worker machine to connect to the main server machine.

Have confirmed:
• there are no firewalls between the machines
• the domain names are pointing to the right machines

• the SSL certificate is working in a single machine configuration

Both machines are running Windows 2012 R2 Standard, in a VM environment. The multiple machine configuration worked fine using FileMaker Server 15. The  certificate is a COMODO Premium SSL Wildcard certificate.

The two machines connected initially using internal IP addresses instead of domain names, and the FileMaker default certificate (using the option Connect using the unverified certificate).

However, using the domain names, with or without the custom SSL certificate, or using the internal IP addresses with the custom SSL certificate, the two machines refuse to connect.

Has anybody else seen this behaviour, can suggest anything else I could try to resolve it?

The system is currently running in a single machine configuration with the custom SSL certificate, but struggles with peak demand.
