I've worked with MODBUS with python (using http://code.google.com/p/modbus-tk/), so in theory you could use pyFileMaker and the python MODBUS libraries to provide the functionality you need without too much difficulty…

There are also MODBUS libraries for PHP (http://code.google.com/p/phpmodbus/) which may suit you better - and would allow the 'supported' FM API to be used - I've just not worked with the PHP libraries, only the Python ones…

You can use Modbus with MBS Plugin and Serial connection.
IP may be done later this year.
I'll check that out. For now, we figured out that the PLC has a server so we can send commands and get a response via a URL. This will work for us initially as we are just using filemaker to monitor, record data and send parameters. If we needed it to directly control the PLC, I think we would got with a serial connection. TCP would be ideal.

This thread cought my eye as Ive been thinking about a similar project but I am still looking for a small inexpensive PLC. Can you say what PLC you are using?
The PLC is called a by a company called TRI, model FMD1616-10
They make a number of different models and using the same program. My brother has been doing all the ladder logic and programing of the PLC and his thinks this is the best for the non-professional.
