There is a hertz rental car on location if you have a license. I do not know about any other shuttle service and searching turns up nothing. You could theoretically take the GO shuttle back to the airport and then from the airport to downtown.
I would call the hotel 1-210-276-2500 to find out if they suggest anything. The hotel might relegate you to using taxis though, a rental car might actually be cheaper than a taxi or a shuttle.
You might also want to post on the bulletin board for a rideshare, I'm sure there will probably be many people with rental cars for the week, or that drove in from out of town (or in town even).
Yeah, these fancy hotels seem to like you stranded. More business for them if you can't get away easily.

I’d offer but I know I’ll be busy Monday morning. Perhaps you could alter your travel plans so you stay overnight in the city center Sunday evening, then you can walk around Monday before taking the ride back to the hotel. The bulletin board isn’t really going full swing on Monday until after registration picks up.
Yes, Lee, the JW Marriott is one of those hotels that tries to keep you there and it is quite a ways (20+ miles) from the big San Antonio downtown tourist attractions like the Riverwalk and Alamo.
The schedule shows Mon., Wed and Thurs evening are FileMaker events with dinner. But Tuesday evening seems to be open for those wanting to explore elsewhere. Since being on the far north side of San Antonio, it might even be easier to do something north of there. Hey, I might even want to drive north to Dripping Springs for the best BBQ in Texas at the Salt Lick (probably an hours drive)!
I think spending a night or several in SA proper, is a nice way to go. In my case, it's virtually a 24 hr travel to get there so I want a bit of recovery time prior to the conference. I thought about staying at the resort the whole time but down town beckoned.
Thus, I'm probably interested in a ride share lateish Monday morning, to transfer to the resort.
I could offer a ride from the city center to JWM on Sunday afternoon, but not Monday.

FYI, if you're renting a car at the airport or driving to the JW Marriott, the parking is free at the hotel. I just phoned and they confirmed no additional cost to park a vehicle.
Is this only for on-site Hertz, or for any rental company? I’m renting with Hertz so that I can return my car and not have to pay for it to sit idle Monday-Thursday.

According to the hotel's website, on-site parking is complimentary. From past travel experiences with both Marriott hotels as well as other hotels with on-site rental companies, this means that any car, regardless of rental company, would be able to park for free.
There is a link to get a discounted shuttle rate from the airport to the JW Marriott on the DevCon website under the Hotel & Travel section. The price is $55 round-trip or $32 one way per person.
I called the hotel this week and asked about ransportation options to the city center and its attractions. From what I was told, the only way to get downtown is to take a taxi or rent a vehicle at the on-site Hertz.
