This would be the correct way, with the default result of "00". Your way would only work if the field was named defaultResult, and the field contained "00"

Unfortunately, that doesn't quite work. It I type anything into the Sample Sub Type field other than Tumour, Adjacent, Healthy or Negative, then Case returns the '00' space-filler, as desired. However if I leave the Sample Sub Type field blank, I get no result. And I'm specifically wanting the '00' space-filler when the Sample Sub Type field has been left blank ...
It should work. Is the calc stored or unstored, and is the result type 'text'? Is the 'do not evaluate if all referenced fields are empty' checked?
The calculation wasn't stored, the result was 'text', and most critically, the 'do not evaluate if all referenced fields are empty' box was checked. I un-checked the latter, it immediately solved it. Woot.
